\nd\u00e9soxyribonucl\u00e9ique), soit de l'ARN (acide ribonucl\u00e9ique), qui sont utilis\u00e9es en m\u00e9decine, en \nrecherche biom\u00e9dicale et en biochimie et chimie analytiques. \nIls se distinguent par leur capacit\u00e9 \u00e0 se lier s\u00e9lectivement et fortement \u00e0 des cibles sp\u00e9cifiques, \nqu'il s'agisse de prot\u00e9ines, de petits compos\u00e9s chimiques ou m\u00eame de cellules.<\/p><p style=\"text-align: left;\">La conception des aptam\u00e8res commence dans le laboratoire de Novaptech, o\u00f9 les chercheurs \nutilisent un processus appel\u00e9 SELEX (Systematic Evolution of Ligands by EXponential \nenrichment) pour isoler les aptam\u00e8res ayant une affinit\u00e9 particuli\u00e8rement \u00e9lev\u00e9e pour la cible \nsouhait\u00e9e. Il s'agit d'un processus complexe en plusieurs \u00e9tapes :<\/p><p style=\"text-align: left;\" class=\"translation-block\">1. S\u00e9lection in vitro : le processus de s\u00e9lection commence par une biblioth\u00e8que al\u00e9atoire \nd'oligonucl\u00e9otides (fragments d'ADN ou d'ARN) contenant des milliards de s\u00e9quences \ndiff\u00e9rentes. Ces s\u00e9quences sont expos\u00e9es \u00e0 la cible que l'on souhaite d\u00e9tecter ou cibler. Les \naptam\u00e8res qui se lient \u00e0 la cible sont alors r\u00e9cup\u00e9r\u00e9s.<\/p><p style=\"text-align: left;\" class=\"translation-block\">2. Amplification : les s\u00e9quences qui se sont li\u00e9es \u00e0 la cible sont amplifi\u00e9es par PCR (r\u00e9action \nde polym\u00e9risation en cha\u00eene) pour produire une plus grande quantit\u00e9 de candidats.<\/p><p style=\"text-align: left;\" class=\"translation-block\">3. R\u00e9p\u00e9tition du processus : ce cycle de s\u00e9lection\/amplification est r\u00e9p\u00e9t\u00e9 plusieurs fois pour \nam\u00e9liorer l'affinit\u00e9 et la sp\u00e9cificit\u00e9 des candidats vis-\u00e0-vis de la cible.<\/p><p style=\"text-align: left;\" class=\"translation-block\">4. Validation : les candidats finaux obtenus sont ensuite test\u00e9s pour confirmer leur capacit\u00e9 \u00e0 \nse lier sp\u00e9cifiquement \u00e0 la cible.
